what should I do now? I paid the item already and the seller is not replying and yesterday, i checked his profile but his name is not a registered user anymore. I got a receipt of my payment but I did not received any SENT NOTIFICATION. please help. Thanks.

Don't worry, you will not lose your money. Go to the item in 'my Ebay' and on the right find on the drop down list 'I didn't receive it' and make a claim stating that the seller is no longer registered. Follow the instructions and you should get your money back shortly.
