they must be listed as Buy it Now and in a category that supports multi choice (eBay do not offer this feature in all categories)

These are the categories that are enabled for variations.
Baby
Cameras
Clothing, Shoes, Accessories
Computers/Tablets & Networking
Crafts
Electronics
Gift Cards
Health & Beauty
Home & Garden
Jewellery & Watches
Lots More...
Phones & Accessories
Sporting Goods
Vehicle Parts & Accessories
If your item doesn't fit into one of these you will not see the option to create variations.

I'm not sure whether the others actually answered your question or not, but if you can't find how to get started, look immediately under the category when you're doing a new listing and before the bit that says 'describe your item' and there'll be bold black writing on a light grey background that says "List multiple variations of your item". Immediately to the left of these words is an arrow - click on this and you'll get more details.

Thanks for your assistance with creating variations with a drop down box. I should have asked how to do it in turbolister. However, I have found that it can be done in ebay advanced,(as you said), and in more categories than are listed in turbolister too. Thankyou for your assistance.
