They won't.
Buyers pay whatever the postage cost or overall cost comes up as in the ad.
Asking for more after a sale is dodgy.
Fact is, a buyer will look at the total cost of an item and make a decision, based on that, if they are willing to buy or not. If you suddenly up the postage by a significant amount (and I presume it must be a significant amount, because you probably wouldn't be worrying about it if we were only talking eg 50c), then a buyer would probably feel that they may not have bought it had they known it was going to be that dear.
That's how I would feel, anyway. I'd be disgruntled. It's not how ebay works.
So you, as seller, have a decision to make. You can either post the item and pay the difference or else you can cancel the sale and refund. If you do the latter, maybe contact the buyer, admit to making a mistake and apologise but say you needed to cancel.
