The buyer selects Buy it now then you mark it as paid and mark it as sent.
If it was sold from elsewhere other than through eBay contact, then you can end item in the drop down menu to the right of item in your selling list.
Keep in mind eBay are sanctioning many members at present for off eBay sales using messages that appear to be suspicious.
