I have had an email from a buyer in Greece who wants to purchase from my eBay store but I only have items listed on Australia eBay.
Can I just change the shipping preferences to accept overseas purchases?

I would ask the buyer which items he wants to purchase and revise those listings to accept worldwide purchases and enter the relevant postage costs. I personally would quote registered mail to cover yourself from any claims of non delivery.
If you haven't sold Internationally before you will also be required to sign ebays International selling agreement.
