I sold an item (rather large and extremely heavy) through buy now. In my listing it clearly states pick up only or buyer to organize courier at own expense, and the town for collection.
The buyer made no previous contact before purchase. Once purchased, I sent a message with instruction of where to collect from, and a range of times. The buyer automatically cancelled the order citing it wont arrive on time.
What are my options regarding this? Is there anyway to penalise the buyer, or do I just lose out on the sale due to this.
Note the buyer has already paid, just refusing to collect.

Only you the seller can cancel the sale, not the buyer. They can ask you to cancel, that's all.
Easiest way to go forward is to cancel the sale using "problem with buyer's address" then you'll get your fees back.
You can't penalise the buyer, just move on and relist. Make sure you block the buyer from dealing with you again though.

The buyer can't cancel, they can ask you to cancel and you can choose to accept or not
And no, you cannot penalise the buyer (unless in the case of a non payment which you can report and they will get a strike from eBay)
You can also put the buyer on your blocked buyers list so they do not do the same to you again

You have the money. You've let the buyer know where and when to pickup. Anything more isn't your problem.

Yes, but the buyer has obviously requested a cancellation.
They are not going to pick up, they are not about to arrange a courier, they want their money back.
If it is those alloy wheels, then $250 is a lot of money.
If the seller just ignores things, the buyer is likely to appeal to ebay or paypal on the basis they did not receive their item (yes, I know it was pick up but we had this happen to us once, where buyer kept changing the pick up date and not turning up then shocked us by opened a claim for item not received and got an instant refund. They had never asked us to cancel)
So they can get the money back anyway, the seller might as well cancel now and at least get the fees refunded.
And no, pinkcookies, you can't penalise the buyer although I know it is really annoying to get buyers like this.

With your experience, I would have expected a factual response.
The buyer allegedly cancelled (an impossibility).
What the OP should do, or have done, is cancel using problem with buyer's address. The buyer gets their money back, the seller gets their fees back (net zero), can relist, and can sleep at night knowing they are not an amoral shyster.
