I have bought and paid for an item on eby via Paypal since 06/10/16 ... after numerous emails to seller I have yet to receive a response. The purchase is for a large amount. Ebay says I have not paid for it, Paypal has deducted the money from my bank but the transaction on Paypal is listed as not claimed yet by seller ... I cannot get any help from Ebay or Paypal as all contact numbers or email options are greyed out ... its quite frustrating and is a rude reminder why I left ebay in the first place ages ago.
What are my options ? Can I cancel the payment ? Does this remove the item from my reminder to pay on ebay ? Its been 12 days and I am 400 bucks down

do you have an estemated date for delivery? you should have. you need to wait until that date is reached then lodge an item not recieved case. then either the seller responds or you get your money back.
i'm taking it you have not recieved an email indicating your item has been posted.
use the 'help & contact' button at the top of the page, follow the links until you get to the phone call bit, then request a call back. explain your problem to the person who calls you.

If the payment is not claimed then there should be a link to cancel it. If their isn't then phone Paypal and ask them to return the funds to you. If the seller does not claim the funds within 30 days of payment then they should automatically go back to you anyway. If you funded the payment with a credit card it will go back onto the card and will take a couple of days longer.
The most likely cause is the seller only has a personal account and would need to upgrade, if it is a large amount I would just get it back as the seller is either trying some type of scam or is very inexperienced, neither scenario is one I would risk my money on.
Do not go through an ebay dispute, their rigid system will still make you wait and jump through hoops that are not needed so stick to dealing with Paypal.
Ebay disputes are only best for not as described cases.

Are you sure the seller is away for the next year? That sounds rather unlikely.
Can you post the item number so someone can have a look for you.
By the way, it is better to start a new thread than drag up an old one which few people will read.
