I just purchased an item. The listing had a postage price. The seller says that the postage price is only an esitamte and does not have to accept it. Is that correct?

Absolute rubbish.
The listed postage cost is the cost that will show on your invoice.
Anything more they are having a 'lend' of you I think.
Certainly question it - if they persist - report them.

If it is a new seller, maybe they are unaware of the procedure, but that is still not an excuse, there are many help options on eBay for a seller to check before they list.
Advise them that you will be paying for the contracted price (item cost and postage as advertised) that is on the invoice.
If they disagree, don't push it, there is no point as you can not force them to send to you, but leave a negative "seller would not honour sale, requested extra postage" sort of thing.
You could pay, using paypal, then go through the INR resolution if the seller refused to send, but that is time wasting and not really worth it.
Keep all messages though, incase seller decides to open NPB.

Personally if the pay now advice shows the correct postage I would pay, using Paypal of course, then the seller has to either post for the agreed price or cop a defect and a neg. If you don't pay then the seller may open an unpaid item dispute and although you would probably get a strike overturned you would not be able to leave the seller the neg they deserve.
Perhaps direct them to this thread so they can see that the experienced sellers here agree with you and not the seller.
