I have had 2 customers today accidently purchase the same item from me twice. They both want to cancel 1, which is fine. I just want to know what happens with the defects and stuff for sellers if we cancel an unpaid order?
Does it still go against our account?

Yes, as mentioned above, no defect if done correctly.
If you haven't done it before, be aware that it is a slow process. eBay like to confirm that buyers have received refunds from cancelled transactions... even when they never paid in the first place.

I always let the customer know that I will do a cancellation request and that they will need to reply to the message they receive from eBay to confirm they wish to cancel the purchase in order to avoid any penalty to either party. I find that if they believe there may be a penalty involved, they nearly always respond and do so promptly. 90% of the time, I get the "You have cancelled an order" message and the appropriate fee refund, within 30 minutes.

You do still have to pay the fees on cancelled orders, it has happened to me. If you open a unpaid item dispute, the fees are returned to you.

I have always had my fees refunded on cancellations. You do have to do it correctly, choose the first option, "Buyer made a mistake or there is a problem with their address". Also, if the buyer doesn't respond to the cancellation request (rare, if you forewarn them that they need to do so), it takes 7-10 days for the request to time out before you get your fees back. The only time you don't get your fees back is if the buyer contests the cancellation.
