Hypothetically, because I dont know exactly what happened in your particular case -
If I bought a top from a seller listed as "NEW", and it turned up with under arm stains, I give the seller a red dot AND Id go through Ebay or Paypal to demand a refund ALSO, and I would make such a fuss they would give me one regardless of their policies.
Not only has the seller lied in their description of it being "new", but they are not including any stains / defects in the decription as well. That is purposely deceptive (or extremely lazy and thoughtless) and disgusting on so many levels.
BIG difference between a piece of clothing thats been worn by a human, and left with UNDER ARM stains, and a couch with a stain from Harvey Norman!

First, happy birthday and a shame this happened. As a buyer I have had clothing not as stated but would have contacted you before a neg. If no response from you or a resolution (if the stains were genuine on a "new" item) then I would have left a neg. No contact is unfair. Policy for a while now is no stains on clothing. The buyer might have known that too.
In terms of what else to do, if you think there is even a chance it is true and you missed it, then double check in strong light. All anyone can do is their best.

Also, did you turn the garment inside out? Could be staining on the inside that didn't show when you were measuring on the outside. Faded fabric is another thing though. Have you considered asking for photos, and if true offering a refund? The buyer might (might) then be inclined to revise feedback.
