Is it possible to block a buyer after they have won an item but not paid? Will it negate that sale?
I know she's going to be trouble and I'd rather not proceed with the sale, but I also don't want to get a defect for a cancellation.
I know I can still block her, it's just this sale I'm concerned about.

Right now pretty much all you can do is wait to see if/not she pays up.
After 4-days if she has not paid then open an unpaid item dispute.
Then after a further 4-days if she still has not paid then close it.
She will be issued a non-payer strike and you will get back your fees.
She also cannot leave feedback after the case is closed.
If she pays during the dispute tiem then you will have to send out her item and take the risk on whatever feedback she chooses to leave you, if any. The UPI case could make her cranky and you could cop RED.
At this point it will be risky to cancel as she has not requested the cancellation in ebay messages.
If you raise a cancellation she has to agree to it.
If she rejects it you will lose your fees.
If she does nothing then after 10-days it will automatically close and you will get your fees back.
If she accepts it then all is good but she can still leave feedback.

You can block and block communication but because you are in a trans with her, it takes some time to take effect.
No it won't negate the current sale but prevent her from buying other items.
